Diversification and shared features of tumor‐binding antibody repertoires in tumor, sentinel lymph node and blood of three patients with breast cancer

Objectives B cell-mediated immunity can be associated with favorable clinical outcomes in cancer patients. However, the mechanism and features of anti-tumor immune responses are not well understood. In particular, how cells expressing tumor-specific antibodies distributed (in tumor vs. circulation) has been defined. Methods We performed an in-depth analysis cell antibody repertoires derived from tumor, sentinel lymph nodes peripheral blood three treatment-naïve patients breast cancer. integrated transcriptional analysis, next-generation sequencing immunoglobulin heavy-chain gene rearrangements phage display to define clonal architecture microenvironment, identify against autologous tissues. Results lineage mapping across libraries generated node revealed that some expanded clones overlap between node, fewer overlapped blood. Notably, tumor-associated or tumor-binding recovered panning tracked back through tissues harboured extensive somatic hypermutations both node. Conclusions These findings suggest evolving humoral response targets possibility monitoring interest after identifying them samples. Studying dynamics specificity may provide insights into characteristics successful immunity, a means for therapy yield novel personalised therapies.
